Transcription keeps it simple

چکیده

new report shows that a eukaryotic transcription factor is directly activated by a metabolite of the pathway it controls. Although this idea has been proposed for several regulatory systems, Christopher Sellick and Richard Reece (University of Manchester, UK) are the first to demonstrate direct binding to and activation of a eukaryotic transcription factor by a small molecule metabolite. The interaction controls the activity of yeast Put3p, an activator of PUT1 and PUT2 transcription. Put1p and Put2p convert proline into glutamate— a high quality source of nitrogen. Put3p binds upstream of the enzymes’ transcription start sites whether or not proline is available, but Sellick and Reece show that waste is avoided by limiting activation to proline-rich conditions. In vitro, Put3p alone was inactive, but transcriptional activation required only the addition of proline. The direct binding of proline to Put3p was weak, probably so the pathway is turned on only when proline is abundant. When preferred nitrogen sources such as glutamine are present, Put3p is inactive even in the presence of proline, suggesting that other mechanisms ensures that proline is converted only when alternative sources are not available.

ELAN development, keeping pace with communities' needs

ELAN is a versatile multimedia annotation tool that is being developed at the Max Planck Institute for Psycholinguistics. About a decade ago it emerged out of a number of corpus tools and utilities and it has been extended ever since. This paper focuses on the efforts made to ensure that the application keeps up with the growing needs of that era in linguistics and multimodality research; growi...

Adaptive Online Learning for Nonstationary Problems

An adaptation algorithm for online training is examined. For stationary tasks it can reduce the learning rate to reach the best convergence. Instead of simple annealing, it keeps the learning rate exible, such that it can also adapt to nonstationary tasks. Di erent tasks, abrupt or gradual changes, and di erent guidance measures are discussed.

Rapid Prototyping with AutoFocus

In most cases, it is simple inconvenience of use that keeps formal methods from being put to industrial use. This paper argues that functionalities, even though of simple formal principles, can be decisive for the applicability of such a formal development tool. Several of those functionalities, as found in the AUTOFOCUS tool prototype, like integrated graphical and hierarchical description tec...

A Q-learning Based Continuous Tuning of Fuzzy Wall Tracking

A simple easy to implement algorithm is proposed to address wall tracking task of an autonomous robot. The robot should navigate in unknown environments, find the nearest wall, and track it solely based on locally sensed data. The proposed method benefits from coupling fuzzy logic and Q-learning to meet requirements of autonomous navigations. Fuzzy if-then rules provide a reliable decision maki...

Rain Initiation Time in Turbulent Warm Clouds

A mean field model is presented that describes droplet growth resulting from condensation and collisions and droplet loss resulting from fallout. The model allows for an effective numerical simulation. The numerical scheme that is conservative in water mass and keeps accurate count of the number of droplets is applied, and the way in which the rain initiation time depends on different parameter...
